The 26-strong burger brand has secured the 1,000 sq ft Southwark Tandoori restaurant site on The Cut, near Waterloo station.
It comes just seven months after the chain opened in the former Feng Sushi site opposite Royal Festival Hall, which is a five minute walk away.
Earlier this year Honest Burgers secured a £17m funding package from its banking partner Santander to help fund its growth pipeline.
The brand launched in Brixton in 2011 and now has 23 London restaurants and one each in Bristol, Cambridge and Reading, with plans to open in Manchester.
Sales at its debut regional site in Cambridge exceeded expectations, giving the group confidence to further expand across the UK.
The company is serious about maintaining its quality as it grows amid a crowded burger market. Last year it invested £600,000 in setting up its own butchery in order to control its supply chain. The patties are now being introduced to menus across all its restaurants.
